In this paper, Stefán Sigurðsson and colleagues studied epigenetic modifications of the BRCA1 gene in breast cancer. BRCA1 was found to be promoter hypermethylated in 3% of breast cancer cases and the methylation status was predictive of improved disease outcome in patients receiving cytotoxic DNA damage-inducing drugs. In this article, Guðrún Valdimarsdottir and colleagues substantiate the importance of BMP9/ALK1 in vascular commitment of hESCs and endothelial sprouting via the secreted extracellular matrix protein EGFL7. They demonstrate the connection of EGFL7 to the NOTCH- and ERK pathway and reveal its role in remodeling of the extracellular matrix using CRISPR/Cas9-mediated deletion of EGFL7.
