Title: Sensitivity to PARP Inhibition in BRCA2 Deficient Cells

Abstract: BRCA2 plays a crucial role in DNA Double Strand Break (DSB) repair. Cells deficient in BRCA2 have impaired ability to repair DSBs and are therefore highly susceptible to DNA damage. PARP1 is a protein responsible for mediating single stranded break repair and has been a target of cancer therapy. By inhibiting PARP1, single stranded breaks may develop into DSBs, potentially killing the cell. We investigated the impact of PARPi on cells containing the Icelandic BRCA2 founder mutation 999del5.
